Skin permeation studies with aspirin were performed in male New Zealand white rabbits (n = 24). The animals were divided into groups according to skin condition: healthy, healthy with inflammation (necrobiotic), psoriasis, and wounds. The receptor fluid was analyzed for aspirin concentration by UV spectrophotometry. In healthy animals, the amount of aspirin permeating the skin was maximal (1.85 micrograms/cm2) at 30 min, and increased significantly with time. The permeation profile of aspirin in healthy animals with inflammation was different from that of the healthy animals. In psoriasis, although the amount of aspirin permeating the skin increased with time, the amount of aspirin in the skin was significantly less (1.35 micrograms/cm2) at 30 min than in the healthy animals. The amount of aspirin in the skin of animals with wounds at 30 min was similar to that of psoriasis animals. The amount of aspirin permeating the skin in the healthy and the healthy with inflammation animals was significantly different (P less than 0.05), but not in the animals with wounds.Spontaneous intracerebral hemorrhage (ICH) is one of the most devastating types of stroke. The clinical data from population based clinical trials indicate that timing of initiation of treatment after ICH is important and the early therapeutic window appears to be very narrow. The overall goal of this proposal is to improve our understanding of the pathophysiology of ICH and to develop novel therapeutic strategies. We hypothesize that the mechanism leading to brain hemorrhage, even in the acute setting is a balance of 2 pathophysiological forces, vessel wall shear stress and a pro-inflammatory milieu. The combination of these forces leads to blood vessels that are not stable and thin, leading to rupture. This leads to hematoma formation, which has the potential to contribute to secondary injury. We propose that the application of an angiogenic growth factor may reduce the hemorrhage size and limit the inflammatory response and that this approach may be most effective when applied as soon as possible after hemorrhage onset. To explore this hypothesis, we have developed a model of ICH in mice.
